E=mc2 is Einstein's formula for his theory of relativity. The reference to Robert is of J Robert Oppenheimer, who led the Manhattan Project. The detonation of the bomb at the Trinity site had one person say, "it looked brighter than a thousand suns". The book of the same name is probably what led to the writing of the song!
